Fabrication, Characterization and in vitro Evaluation of Prednisolone Sustained Release Multiparticulate System for Colonic Targeting

Background: Prednisolone (PRD) is orally prescribed for inflammatory bowel syndrome (IBS) as the upper GIT main site of absorption; therefore, long-term PRD dosing decreases therapeutic effectiveness through systemic side effects. Objective: This work focused on formulating sustained-release alginate beads a multiparticulate system colon targeting using prednisolone to be filled in an HPMC capsule. Methods: were prepared by ionotropic gelation technique sodium primary polymer and inulin, guar gum, pectin secondary polymers. In addition impact type quantity, other factors investigated: The CaCl2 concentration tween 80 Thirteen formulations successfully prepared, their properties, such bead size, morphology, percentage encapsulation efficiency, yield, DL, vitro release study buffer media, IBS SEM, FTIR, assessed. Results: showed that close size was not obstacle loading capsules. Further, yield%, EE%, DL% increased according bead’s content increase. Conclusions: optimum formula F3 coated capsules with Eudragit S-100, which gave sustained profiles simulating F13 could last different pH 1.2, 6.8, 7.4.
